    A Platelet/CMC coupled with offline UPLC-QTOF-MS/MS for screening antiplatelet activity components from aqueous extract of Danshen[Pubmed: 26355772]
    Abstract Platelets play crucial roles in thromboembolic and cardiovascular disease. The main platelets membrane receptors include adenosine diphosphate receptors, thrombin receptors, thromboxane prostanoid receptors and collagen receptors. In this study, a Platelet/CMC coupled with offline UPLC-QTOF-MS/MS system was built to screen antiplatelet activity components from aqueous extract of Danshen, which serve as an agent of antiplatelet aggregation in Traditional Chinese Medicine. Rosmarinic acid, lithospermic acid, salvianolic acid B, two isomers of salvianolic acid B, salvianolic acid C, Salvianolic acid D and salvianolic acid H/I were identified as the potential antiplatelet activity components. Moreover, rosmarinic acid, lithospermic acid, salvianolic acid B, salvianolic acid C and danshensu were tested in platelet aggregation in vitro assay. The results suggested their retention time was closely related to the antiplatelet aggregation activities. This study provides a rapid, effective and novel method for screening the potential antiplatelet activity components from Chinese herb medicines.     Pharmacokinetic study of salvianolic acid D after oral and intravenous administration in rats[Reference: WebLink]

    METHODS AND RESULTS:
    A sensitive, specific and rapid LC-MS method was developed and validated for the determination of Salvianolic acid D (SalD) in rat plasma. This method used a single quadrupole mass spectrometer with an electrospray ionization (ESI) source. A single ion monitoring scanning (SIM) mode was employed. It showed good linearity over the concentration range from 3.3 to 666.7 ng/mL for the determination of Salvianolic acid D. The R.S.D.% of intra-day and inter-day precision values were no more than 7.69%, and the accuracy was within 91%−104% at all quality control levels.
    CONCLUSIONS:
    This LC-MS method was applied to the pharmacokinetic study of Salvianolic acid D in rats. A two-compartmental model analysis was employed. The bioavailability of Salvianolic acid D was calculated to be 4.159%±0.517%.

    Isolation and Identification of Salvianolic Acid D and Simultaneous Determination of 4 Salvianolic Acids in Danshen Infusion[Reference: WebLink]
    To establish the method for the isolation and identification of Salvianolic acid D and simultaneous determination of tanshinol,protocatechuic aldehyde,Salvianolic acid D and B in Danshen infusion.
    METHODS AND RESULTS:
    RP-HPLC method was adopted for isolation and content determination.The determination was performed on Kromasil C18 column with mobile phase consisted of acetonitrile-1% glacial acetic acid at the flow rate of 1.0 mL·min-1.The injection volume was 10 μL and column temperature was 30 ℃.The detection wavelength was set at 281 nm. Salvianolic acid D had been succeddfully isolated and indentified.The linearities of tanshinol,protocatechuic aldehyde,Salvianolic acid D,salvianolic acid B were 60.9~975.0,8.7~139.0,11.0~175.8,2.7~43.2 μg·mL-1.The average recoveries rate were 99.1%,100.2%,98.3%,100.4%,102.5%,101.1%,101.4%,97.0%,102.1%,100.6%,99.2%,101.3%,and the RSDs were 0.94%,0.97%,2.78%,1.07%.
    CONCLUSIONS:
    The method is simple,rapid and specific.It may be suitable for the quality control of Danshen infusion.
