Lobetyolin has a protective role in gastric mucosa injury, and the mechanism may be relate to the increase in level of 6-K-PGF1αand the inhibition the excretion of gastric acid and EGF. Lobetyolin inhibits the gene expression of MUC5AC mucin induced by PMA.

    Effects of Lobetyolin, Lobetyol and Methyl linoleate on Secretion, Production and Gene Expression of MUC5AC Mucin from Airway Epithelial Cells.[Pubmed: 25473407]
    In this study, we investigated whether Lobetyolin, lobetyol, and methyl linoleate derived from Codonopsis pilosula affect MUC5AC mucin secretion, production, and gene expression from airway epithelial cells.
    METHODS AND RESULTS:
    Confluent NCI-H292 cells were pretreated with Lobetyolin, lobetyol, or methyl linoleate for 30 minutes and then stimulated with phorbol 12-myristate 13-acetate (PMA) for 24 hours. The MUC5AC mucin gene expression, and mucin protein production and secretion were measured by reverse transcription polymerase chain reaction and enzyme-linked immunosorbent assay, respectively. Lobetyolin, lobetyol, and methyl linoleate inhibited the gene expression of MUC5AC mucin induced by PMA; Lobetyolin did not affect PMA-induced MUC5AC mucin production. However, lobetyol and methyl linoleate inhibited the production of MUC5AC mucin; Lobetyolin and lobetyol did not significantly affect PMA-induced MUC5AC mucin secretion from NCI-H292 cells. However, methyl linoleate decreased the MUC5AC mucin secretion.
    CONCLUSIONS:
    These results suggest that among the three compounds, methyl linoleate can regulate gene expression, production, and secretion of MUC5AC mucin by directly acting on the airway epithelial cells.

    Protective Effect of Lobetyolin on Gastric Mucosa of Experimental Gastric Ulcer in Rats[Reference: WebLink]
    To study the protective effect of Lobetyolin on gastric mucosa of experimental gastric ulcer induced by ethanol in rats.
    METHODS AND RESULTS:
    The gastric ulcer model was established in rats by inducing ethanol.The index of ulcer was measured;the levels of Gas and 6-K-PGF1αwere examined by the method of radioimmunity;the level of EGF was measured by ELISA.As compared with model group,in Lobetyolin small dose group,the index of ulcer was markedly decreased,the level of Gas was decreased while the level of 6-K-PGF1αwas increased.In the degree,the level of EGF was increased.
    CONCLUSIONS:
    Lobetyolin has a protective role in gastric mucosa injury,and the mechanism may be relate to the increase in level of 6-K-PGF1αand the inhibition the excretion of gastric acid and EGF.

    METHODS AND RESULTS:
    The compounds were isolated and purified by column chromatography and their structures were elucidated through spectroscopic techniques (NMR) and physicochemical properties. The compounds were isolated as hesperidin(I), n-hexyl beta-sophoroside(II), atractylenolide III (III), Lobetyolin(IV), Lobetyolinin(V), taraxerol(VI), taraxeryl acetate(VII), alpha-spinasterol(VIII),9,10,13-trihydroxy-(E)-11-octadecenoic acid (IX),beta-sitosterol(X),beta-daucosterol(XI)and sugar(XII).
    CONCLUSIONS:
    Compounds 1 -2 and 9 are isolated from this plant for the first time.
